Vitamin D3 Controls TLR4- and TLR2-Mediated Inflammatory Responses of Endometrial Cells

چکیده
Objectives: Vitamin D has potent immunoregulatory features and modulates innate adaptive immune responses. There is a significant association between intrauterine infection-associated inflammatory responses pregnancy complications such as abortion preterm labor. Here, we investigated how 1,25 (OH)2 D3 could modulate of endometrial cells. Design: This an in vitro experimental study. Endometrial stromal cells (ESCs) whole (WECs) were collected from 15 apparently normal women, the immunomodulatory effects on lipopolysaccharide (LPS)- or lipoteichoic acid (LTA)-treated ESCs WECs investigated. Participants/Materials, Setting, Methods: Women with no history abortion, infertility, endometriosis, sign vaginal infection enrolled this samples by gynecologists using Pipelle pipette proliferative phase menstrual cycle. treated either LPS LTA. The levels IL-6, IL-8, TNF-α culture supernatants quantified ELISA technique. TLR2, TLR4, MyD88 expressions assessed RT-qPCR. TLR4 expression at protein level was studied Western blot Results: Dihydroxycholecalciferol (1,25 D3) significantly reduced production LPS-activated IL-6 LTA-stimulated WECs. In contrast, pretreatment increased IL-8 LPS- markedly LPS-induced ESCs. treatment induced gene expression. effect reversed when these pretreated before stimulation LPS. Limitations: Because small size samples, doing experiments all together some not feasible. Confirmation results obtained here needs well-designed vivo studies. Conclusions: molecule essential for maintaining homeostasis controlling potentially harmful associated female reproductive tract infections.

lipopolysaccharide- and lipoteichoic acid-mediated pro-inflammatory cytokine production and modulation of tlr2, tlr4 and myd88 expression in human endometrial cells
background: toll-like receptor (tlr)-mediated inflammatory processes are supposed to be involved in pathophysiology of spontaneous abortion and preterm labor. here, we investigated functional responses of human endometrial stromal cells (escs) and whole endometrial cells (wecs) to lipopolysaccharide (lps) and lipoteichoic acid (lta). methods: endometrial tissues were obtained from 15 cycling wo...
متن کاملCholesterol crystals enhance TLR2- and TLR4-mediated pro-inflammatory cytokine responses of monocytes to the proatherogenic oral bacterium Porphyromonas gingivalis
Cholesterol deposits and pro-inflammatory cytokines play an essential role in the pathogenesis of atherosclerosis, a predominant cause of cardiovascular disease (CVD). Epidemiological evidence has linked periodontal disease (PD) with atherosclerotic CVD. Accordingly, viable periodontal pathogens, including Porphyromonas gingivalis, have been found in atherosclerotic plaques in humans and mice. ...

متن کاملTLR4- and TLR2-mediated B cell responses control the clearance of the bacterial pathogen, Leptospira interrogans.
Leptospirosis is a widespread zoonosis caused by pathogenic Leptospira interrogans that are transmitted by asymptomatic infected rodents. Leptospiral lipoproteins and LPS have been shown to stimulate murine cells via TLRs 2 and 4. Host defense mechanisms remain obscure, although TLR4 has been shown to be involved in clearing Leptospira. In this study, we show that double (TLR2 and TLR4) knockou...
متن کاملEstrogen controls vitamin D3-mediated resistance to experimental autoimmune encephalomyelitis by controlling vitamin D3 metabolism and receptor expression.
Multiple sclerosis (MS) is an autoimmune, neurodegenerative disease with a rapidly increasing female gender bias. MS prevalence decreases with increasing sunlight exposure, supporting our hypothesis that the sunlight-dependent hormone 1,25-dihydroxyvitamin D(3) (1,25-(OH)(2)D(3)) is a natural inhibitor of autoimmune T cell responses in MS. We found that vitamin D(3) inhibited experimental autoi...
